JC-1 works as a mitochondrial potential indictor. In intact mitochondria with membrane potential, the polymeric form of JC-1 at high concentration shows red fluorescence excited at 590 nm.

Store in dark at -20 ℃.
使用方法
General protocol: 1. Prepare a 1mg/mL stock solution of JC-1 in DMSO. Protect from light, and store at –20˚C. 2. Prepare a small aliquot of fresh 10 µg/mL JC-1 in the appropriate cell culture medium with 10% calf serum at 37˚C. Note: The working solution of JC-1 should be used immediately after it done. Discard staining solutions that are older than 3 min. 3. Aspirate the cell culture medium from cells grown on a coverslip. Do not allow the cells to dry out. 4. Apply 50 µL of the JC-1-labeling solution to the cells. 5. Incubate the cells with the JC-1 labeling solution in a covered dish for 10 min at 37˚C in a cell culture incubator. 6. Aspirate the labeling solution, and rinse the cells three times in cell culture medium without JC-1. 7. Mount the coverslips. 8. Image the cells. For the JC-1 monomeric dye, λex maximum is 514 nm and λem maximum is 529 nm. For the J-aggregate form of JC-1, λex maximum 485–585 nm and λem maximum 590 nm.{

JC-1 works as a mitochondrial potential indictor. In intact mitochondria with membrane potential, the polymeric form of JC-1 at high concentration shows red fluorescence excited at 590 nm. During cell apoptosis, it leaks from the depolarized mitochondria and shows green fluorescence excited at 529 nm. Thus, the ratio between red and green fluorescence of cells loaded with JC-1 is used to detect cell mitochondrial potential.
