LIN28 inhibitor LI71 is a potent and cell-permeable LIN28 inhibitor, which abolishes LIN28-mediated oligouridylation with an IC50 of 7 uM. LIN28 inhibitor LI71 directly binds the cold shock domain (CSD) to suppress LIN28's activity against let-7 in leukemia cells and embryonic stem cells.

描述LIN28 is an RNA-binding protein that regulates the maturation of the let-7 family of microRNAs by bipartite interactions with let-7 precursors. Through inhibition of let-7 biogenesis, LIN28 functions as a pluripotency factor, as well as a driver of tumorigenesis. LIN28 inhibitor LI71 is a potent and cell-permeable inhibitor of LIN28, which inhibits the interactions between LIN28 and the let-7 family with IC₅₀s of ~7 and ~27 μM in fluorescence polarization assay and in oligouridylation assay, respectively. LI71 could significantly reduce the relative Renilla luciferase activity in cells expressing LIN28A and LIN28B at 50-100 μM. Moreover, treatment of DKO+A cells with 100 μM LI71 for 48 hr increased levels of mature let-7 species compared to vehicle-treated cells^[1].
