描述DS-3032b is an orally available MDM2 (murine double minute 2) antagonist with potential antineoplastic activity. DS-3032b binds to, and prevents the binding of MDM2 protein to the transcriptional activation domain of the tumor suppressor protein p53. DS-3032b is 10-fold more potent than the first-generation inhibitor nutlin-3a. DS-3032b treatment enhanced TP53 target gene expression and induced G1 cell cycle arrest, senescence and apoptosis. CRISPR-mediated MDM2 knockout in neuroblastoma cells mimicked DS-3032b treatment.
