ZXH-3-26 is a BRD4 heterobifunctional small-molecule ligand (PROTAC), shows activity exclusively on the BRD4 BD1 (DC50/5h=5 nM) and spares degradation of BRD2 or BRD3 in cellular degradation assays; ZXH-3-26 represents the first small molecule to allow pharmacologic targeting of BRD4 without significant inhibition or degradation of BRD2 and BRD3.

描述ZXH-3-26 is a potent and highly selective PROTAC targeting on BRD4_BD1 with DC50 value of ~ 5nM post 5-hour treatment in cells stably expressing BRD4_BD1-EGFP. Compared with the best pan-BRD degrader dBET6, ZXH-3-26 exhibited higher selectivity and almost completely degraded endogenous BRD4 at concentration ranging in 10-1000nM in HEK293T cells post 5-hour treatment, with no obvious change of BRD2 or BRD3.
