使用方法
Protocol for in vivo labeling of cortical astrocytes with sulforhodamine 101 (SR101)ACSF (Artificial Cerebrospinal Fluid) prepare:125 mM NaCl5 mM KCl10 mM D-glucose10 mM HEPES2 mM MgSO42 mM CaCl2Adjust pH to ~7.3 with NaOH. Osmolarity should be ~300.For Topical Application:1. Prepare SR101 solution (typically 200–300 µM in ACSF) either fresh or from a stock solution. 2. Perform a craniotomy above the brain region of interest. Carefully perforate the dura locally to allow dye penetration into cortical tissue. Do not remove the dura unless absolutely necessary. If dura removal is required, perform SR101 staining before the dura removal. To perforate the dura, use a stereomicroscope at high magnification and ample ACSF immersion. Use the tip of a new hypodermic needle (e.g., 27- or 30-gauge) to make a small (~0.5-mm diameter) incision away from surface blood vessels near one edge of the craniotomy.
