FITC (Fluorescein Isothiocyanate) is a yellow-orange powder with a maximum excitation wavelength of 494 nm. Once excited, at maximum emission, it shows yellow-green fluorescence at a wavelength of 520 nm. It has high absorption rate, excellent fluorescence quantum yield and good water solubility, and is one of the most widely used green fluorescein derivatives in biology. Its isothiocyanate group can react with the amino terminal or primary amine of the protein which make it can be used for protein labeling including antibodies and lectins. In addition to being used as a protein marker, it can also be used as a protein fluorescent tracer, labeling antibodies for rapid identification of pathogens and for micro-sequencing of proteins and peptides (HPLC).
