使用方法
Detection of cervical cancer cells in human blood samples 1. Human blood samples are mixed with signal-cell suspension of cervical cancer cells pre-labeled with DAPI. 2. IR-783 dye (20 μM) is added, incubation at 37°C for 30 min. 3. The mononuclear and cancer cells are isolated form mixture via the gradient centrifugation method using Histopaque-1077. 4. After fixation of normal and cancer cells, the mixture is observed under confocal microscope to identify the cancer cells via the positive staining. Flow cytometry is used to count for the total number of cancer cells in the blood. IR-783 dye accumulation in tumors of live mice Mice are injected with IR-783 dye at a dose of 0.35 mg/kg when tumor size reached ~2-8 mm in diameter, as assessed by X-ray and by palpation. IR-783 can be used as a starting material in the synthesis of IR-808 dye for biological applications. It can also be used in the formation of NIR fluorescent human serum albumin nanoparticles for the detection of a tumor. It finds potential usage as a contrasting agent for the multimodal in vivo imaging. Excitation wavelength ~633 nm, emission ~780 nm.
