5-Ethynyluridine is used to help detect RNA synthesis in cells by its biosynthetic incorporation into newly transcribed RNA. Once fed to cells, it is incorporated during active RNA synthesis and can then be detected using a copper-catalyzed click reaction with fluorescent dyes. The cells can then be analysed by fluorescence imaging, flow cytometry or high content imaging analysis.

描述5-Ethynyluridine is a cell-permeable uridine analog. 5-Ethynyluridine can be used in vitro and in vivo to label newly synthesized RNA. 5-Ethynyluridine can be used to study changes in transcription since it can detect global RNA transcription temporally and spatially in both in-vitro and in-vivo with the modified nucleotide. In vivo studies in mice, EU was injected intraperitoneally (330–750 μmol/g animal), resulting in labelling of nascent RNA in cells of multiple organs and tissues, but not in the central nervous system^[1]. The resulting ethynyl-functionalized RNA can subsequently be detected via Cu(I)-catalyzed click chemistry that offers the choice to introduce a Biotin group (via Azides of Biotin) for subsequent purification tasks or a fluorescent group (via Azides of fluorescent dyes) for subsequent microscopic imaging^[2].
Presolski et al.^[3] and Hong et al.^[4] provide a general protocol for Cu(I)-catalyzed click chemistry reactions that may be used as a starting point for the set up and optimization of individual assays.
