描述Naxitamab is a humanised (IgG1) anti-GD2 (hu3F8) monoclonal antibody was developed by the Memorial Sloan Kettering Cancer Center (with commercial rights licenced to Y-mAbs therapeutics Inc.) for the treatment of neuroblastoma, osteosarcoma and other GD2-positive cancers. Naxitamab (hu3F8-IgG1) had kon and koff values of 9.19 and 1.03E, respectively and a KD of 11 nmol/L by surface plasmon resonance for binding to GD2 coated onto CM5 chips. Cross-reactivity with gangliosides GM2, GD1a, GD1b, GT1b, GD3 and GQ1b was low (0, 0, 4.5, 0, 0 and 0% respectively). Naxitamab (hu3F8-IgG1) demonstrated direct cytotoxicity against neuroblastoma cell line LAN-1 (EC50 5.1 μg/mL) in vitro. In antibody-dependent cell-mediated cytotoxicity assays using peripheral blood mononuclear cells (PBMC) and polymorphonuclear leukocytes (PMN) from volunteers as effectors and neuroblastoma LAN-1 cells as targets the potency of naxitamab (hu3F8-Ig1) relative to murine (mu3F8) was 217 with PBMC, 19 with PMN 12 with NK-92MI-CD16, 15 with NK-92MI-CD32 and 0.40 with complement mediated cytotoxicity.
